In vitro study | SP600125 is an ATP-competitive c-Jun N-terminal kinase (JNK) selective inhibitor with an IC50 of 40 nM to 90 nM. SP600125 acts on Jurkat T cells and inhibits c-Jun phosphorylation with an IC50 of 5 μm to 10 μm. SP600125 acts on CD4 However, later studies have shown that SP600125 also inhibits the aromatic hydrocarbon receptor (AhR) Mps1, and A series of other serine/threonine kinases, including Aurora kinase A, FLT3, MELK, and TRKA. 20 μm SP600125 acts on mouse beta cells MIN6 to induce p38 MAPK phosphorylation and its downstream CREB-dependent promoter activation. 20 μm SP600125 acts on HCT116 cells to stop mitosis at G2 phase and induce intranuclear replication. |
In vivo study | SP600125 at 15 mg/kg or 30 mg/kg significantly inhibited lipopolysaccharide (LPS)-induced TNF-α expression and CD4 induced by CD3 antibody in mice |

biological activity | SP600125 (Nsc75890) is a broad-spectrum JNK inhibitor that acts on JNK1, JNK2 and JNK3. IC50 in cell-free test is 40 nM, 40 nM and 90 nM respectively, which is 10 times higher than that on MKK4 and 25 times higher than that on MKK3, MKK6, PKB and PKCα, it is 100 times more selective than ERK2, p38, Chk1, EGFR, etc. SP600125 is also a broad-spectrum serine/threonine kinases inhibitor, including Aurora kinase A, FLT3 and TRKA, with corresponding IC50 values of 60 nM, 90 nM and 70 nM. SP600125 can inhibit autophagy and activate apoptosis. |
